Soils represent the major source of the atmospheric greenhouse gas nitrous oxide (N2O) and there is a need to better constrain the total global flux and the relative contribution of the microbial source processes. The aim of our study was to determine variability and control of the isotopic fingerprint of N2O fluxes following NH4+-fertilization and dominated by nitrification. We conducted a microcosm study with three arable soils fertilized with 0–140 mg NH4+–N kg−1. Fractions of N2O derived from nitrification and denitrification were determined in parallel experiments using the 15N tracer and acetylene inhibition techniques or by comparison with unfertilized treatments. Soils were incubated for 3–10 days at low moisture (30–55% water-filled pore space) in order to establish conditions favoring nitrification. Dual isotope and isotopomer ratios of emitted N2O were determined by mass spectrometric analysis of δ18O, average δ15N (δ15Nbulk) and 15N site preference (SP = difference in δ15N between the central and peripheral N positions of the asymmetric N2O molecule). N2O originated mainly from nitrification (>80%) in all treatments and the proportion of NH4+ nitrified that was lost as N2O ranged between 0.07 and 0.45%. δ18O and SP of N2O fluxes ranged from 15 to 28.4‰ and from 13.9 to 29.8‰, respectively. These ranges overlapped with isotopic signatures of N2O from denitrification reported previously. There was a negative correlation between SP and δ18O which is opposite to reported trends in N2O from denitrification. Variation of average 15N signatures of N2O (δ15Nbulk) did not supply process information, apparently because a strong shift in precursor signatures masked process-specific effects on δ15Nbulk. Maximum SP of total N2O fluxes and of nitrification fluxes was close to reported SP of N2O from NH4+ or NH2OH conversion by autotrophic nitrifiers, suggesting that SP close to 30‰ is typical for autotrophic nitrification in soils following NH4+-fertilization. The results suggest that the δ18O/SP fingerprint of N2O might be used as a new indicator of the dominant source process of N2O fluxes in soils.  相似文献

A 28-day incubation experiment at 12°C was carried out on the decomposition of maize leaf litter to answer the questions: (1) Is the decomposition process altered by chemical manipulations due to differences in the colonization of maize leaf litter? (2) Do organisms using this maize material contribute significantly to the soil microbial biomass? The extraction of the maize straw reduced its initial microbial biomass C content by 25%. Fumigation and extraction eliminated the microbial biomass by 88%. In total, 17% of added maize straw C was mineralized to CO2 during the 28-day incubation at 12°C in the treatment with non-manipulated straw. Only 14% of added C was mineralized in the treatment with extracted straw as well as in the treatment with fumigated and extracted straw. The net increase in microbial biomass C was 79 μg g?1 soil in the treatment with non-manipulated straw and an insignificant 9 μg g?1 soil in the two treatments with manipulated straw. However, the net increase did not reflect the fact that the addition of maize straw replaced an identical 58% (≈180 μg g?1 soil) of the autochthonous microbial biomass C3-C in all three straw treatments. In the two treatments with manipulated straw, the formation of maize-derived microbial biomass C4-C was significantly reduced by 25%. In the three straw treatments, the ratio of fungal ergosterol-to-microbial biomass C ratio showed a constant 60% increase compared to the control, and the contents of glucosamine and muramic acid increased by 18%. The average fungal C/bacterial C ratio was 3.6 in the soil and 5.0 in the recovered maize straw, indicating that fungal dominance was not altered by the initial chemical manipulations of the maize straw-colonizing microorganisms.  相似文献

The antioxidant capacity of methanol and ethanol seed extracts from Euterpe oleracea Mart. (a?aí) against the reactive oxygen species (ROS) peroxyl radicals, peroxynitrite, and hydroxyl radicals was studied with the total oxidant scavenging capacity (TOSC) assay in a modified and automated version. Cold methanol digestion was the most efficient extraction method with respect to the antioxidant capacity. The extracts exhibit good antioxidant capacity against peroxyl radicals, similar to the capacity of the pulp. The antioxidant capacity against peroxynitrite and hydroxyl radicals is even higher. The main antioxidants identified by HPLC-MS and HPLC-CEAD are five different procyanidins (di- through pentamers); furthermore, protocatechuic acid and epicatechin were identified as minor compounds. Determination of TOSC values of HPLC seed extract fractions indicates that the procyanidins contribute substantially to the overall antioxidant capacity. In addition, however, other compounds that have not yet been identified are responsible for a large part of the observed antioxidant capacity.  相似文献

Field experiments show that wheat grown after oilseed rape (OSR) achieves higher yield levels, while the nitrogen (N) application is reduced. However, field experiment data are based on few locations with optimised management. We analysed a large dataset based on farm data to assess the true extent of break crop benefits (BCB) for yield and N fertilisation within German commercial farming.

Across all German states and years, average yield of wheat preceded by OSR was 0.56 Mg ha?1 higher than yield of wheat preceded by cereals (7.09 Mg ha?1), although considerable variation between regions was observed. Mean N application across all states to wheat after OSR was 5 kg ha?1 lower than to wheat after cereals. Choice of wheat types for different end uses (bread flour or animal feed) showed higher (0.77 Mg ha?1) or lower (0.44 Mg ha?1) BCB for yield of wheat cultivated after OSR compared with after cereals. The calculated BCB for yield and N fertilisation were lower than expected from dedicated field experiments and fertiliser recommendations. Thus the advantages of OSR as a preceding crop are generally utilised by commercial farmers in Germany but there is room for improvement.  相似文献

Saline melt water from road salt applications that has percolated into a fine sandy soil in winter is rinsed out of the soil by infiltrating rainwater in the following warmer seasons. This sequence of saturated and unsaturated flow processes associated with saline water transport in a fine sandy soil was studied by simulation and exploratory laboratory experiments. Experiments in soil columns of 300-μm sand revealed that two rinses of pure water, each of one pore volume, were sufficient to reduce the salt concentration by 99% of its original value in the soil column. Simulated time variations of salt concentration in the effluent from the column agreed with experimental results. Based on simulated and experimental results, a sandy soil must become saturated to experience pore water flow in order to efficiently rinse saline snowmelt water. Depending on the saturated hydraulic conductivity and the soil depth, days, weeks, or months of freshwater infiltration in summer are needed to rinse saline melt water from an unsaturated sandy soil after road salt applications in winter. This explains findings of significant salt concentrations in surface and shallow groundwater during summer months, long after road salt application and infiltration has ceased.  相似文献

Nmim content in the soil, N-fertilization and N uptake of winter wheat in the international organic nitrogen long-term fertilization experiment (IOSDV) Berlin-Dahlem During the 9th and 10th year of the long-term IOSDV field experiment micro plots were put in three treatments. Labelled15 N (160 resp. 110 kg/ha N as ammonium sulfate) was fertilized to winter wheat subdivided into three portions. Nmin in soil was determined five times during the season, plant biomass was harvested at different growth stages and N uptake was calculated. Using the15 N-technique permitted a discrimination between fertilizer-N and soil-N. Preferential uptake of fertilizer-N by the wheat crop but also immobilisation in soil were observed until June. Subsequently the immobilized N was remineralized and assimilated by wheat. But the native Nmin of soil was minimaly utilized during the initial growth of wheat. Therefore the absolute amount of soluble N temporarily increased caused by a mineralisation of the organic matter.  相似文献

Chemical investigation of the ethyl acetate extract from the marine-derived Streptomyces sp. isolate B1848 resulted in three new isoquinolinequinone derivatives, the mansouramycins E–G (1a–3a), in addition to the previously reported mansouramycins A (5) and D (6). Their structures were elucidated by computer-assisted interpretation of 1D and 2D NMR spectra, high-resolution mass spectrometry, and by comparison with related compounds. Cytotoxicity profiling of the mansouramycins in a panel of up to 36 tumor cell lines indicated a significant cytotoxicity and good tumor selectivity for mansouramycin F (2a), while the activity profile of E (1a) was less attractive.  相似文献
